Abstract : In the present study, we examined the effects of FFAs on insulin sensitivity and signaling cascades in the C2C12 skeletal muscle cell culture system. We compared the effects of a monounsaturated fatty acid, oleate (C18:1) and a saturated fatty acid, palmitate (C16:0), on insulin sensitivity in the myotube muscle cells. Our data clearly has manifested, the inhibitory effects of PKC on insulin signaling may at least in part be explained by the serine/therionine phosphorylation of IRS-1, which has been implicated in the inhibition of its tyrosine phosphorylation. Both oleate and palmitate treatment were able to increase the serine 307 phosphorylation for IRS-1 as a possible novel mechanism due to the PKC activation as seen in our earlier reports. IRS-1 seine 307 is inducible and responsible for the inhibition of IRS-1 function can be phosphorylated by either IKK or c-jun N-terminal kinase (JNK) as seen in our proteomic kinases screen. Further, another supportive evidence to our finding has manifested that the two FFAs are to activate the IKK/, the stress kinases S6 kinase p70 (p70SK), stress activated protein kinase (JNK) (SAPK), and p38 MAP Kinase (p38MAPK). These data suggest that oleate, may directly induce muscle insulin resistance via PKC-NFB pathway that induce IRS-1 seine 307 and desensitize the PI3K pathways at a number of key targets in the insulin cascade. In the contrary palmitate induces insulin resistance mechanistically through PKC activation, the inhibition of PKB phosphorylation and the IRS-1 seine 307 phosphorylation seen in our data.
